  WB analysis using 83194-4-RR (same clone as 83194-4-PBS)

  Various lysates were subjected to SDS PAGE followed by western blot with 83194-4-RR (HIRA antibody) at dilution of 1:10000 incubated at room temperature for 1.5 hours. This data was developed using the same antibody clone with 83194-4-PBS in a different storage buffer formulation.

  FC experiment of K-562 using 83194-4-RR (same clone as 83194-4-PBS)

  1x10^6 K-562 cells were intracellularly stained with 0.25 ug HIRA Recombinant antibody (83194-4-RR, Clone:240006F8) and CoraLite®488-Conjugated Goat Anti-Rabbit IgG(H+L) (SA00013-2)(red), or 0.25 ug Rabbit IgG Isotype Control RecAb (98136-1-RR, Clone: 240953C9) (blue). Cells were fixed and permeabilized with Transcription Factor Staining Buffer Kit (PF00011). This data was developed using the same antibody clone with 83194-4-PBS in a different storage buffer formulation.

  Affinity and Kinetic Characterization of 83194-4-RR

  Biolayer interferometry (BLl) kinetic assays of 83194-4-RR against Human HIRA were performed. The affinity constant is below 1 pM.

Informations générales

HIRA protein is encoded by a gene within a region of human chromosome 22q11.2 deleted in most patients with the DiGeorge syndrome, a developmental disorder, and was named for its amino acid sequence homology to two S. cerevisiae proteins, Hir1p and Hir2p. HIRA is critical in a specific chromatin assembly pathway that takes place independently of DNA synthesis (PMID: 12049744). HIRA complex associates with various proteins, such as transcription factors, RNA pol II, Prohibitin (PHB), and replication protein A (RPA) suggests that the targeting of the complex at specific locations may exploit additional structural properties and mechanisms (PMID: 30082790).
