  WB analysis using 81820-2-RR (same clone as 81820-2-PBS)

  Various lysates were subjected to SDS PAGE followed by western blot with 81820-2-RR (NR1H4 antibody) at dilution of 1:5000 incubated at room temperature for 1.5 hours. This data was developed using the same antibody clone with 81820-2-PBS in a different storage buffer formulation.

  WB analysis of HepG2 using 81820-2-RR (same clone as 81820-2-PBS)

  WB result of NR1H4 antibody (81820-2-RR; 1:3000; incubated at room temperature for 1.5 hours) with sh-Control and sh-NR1H4 transfected HepG2 cells. This data was developed using the same antibody clone with 81820-2-PBS in a different storage buffer formulation.

  FC experiment of HepG2 using 81820-2-RR (same clone as 81820-2-PBS)

  1x10^6 HepG2 cells were intracellularly stained with 0.25 ug NR1H4 Recombinant antibody (81820-2-RR, Clone:4B4) and CoraLite®488-Conjugated Goat Anti-Rabbit IgG(H+L) (SA00013-2)(red), or 0.25 ug Rabbit IgG Isotype Control Recombinant Antibody (98136-1-RR, Clone: 240953C9) (blue). Cells were fixed and permeabilized with Transcription Factor Staining Buffer Kit (PF00011). This data was developed using the same antibody clone with 81820-2-PBS in a different storage buffer formulation.

  Affinity and Kinetic Characterization of 81820-2-RR

  Biolayer interferometry (BLl) kinetic assays of 81820-2-RR against Human NR1H4 were performed. The affinity constant is 0.807 nM.

Informations générales

Nuclear Receptor subfamily 1, group H, member 4 (NR1H4, also known as FXR) is a receptor for bile acids and has an important role in regulating energy metabolism in liver, muscle and adipose tissues in humans and animals. NR1H4 is highly expressed in liver and has a role in lipid metabolism, whereas none of the other protein-coding genes in the region has known biological links with cholesterol, further supporting a role for NR1H4 in regulation of cholesterol levels. NR1H4 have four isoforms, i.e., FXRalpha1, FXRalpha2, FXRbeta1, and FXRbeta2, and each isoform has a different expressional level and transcriptional activity depending on its location within specific tissues.
