  WB analysis using 82894-7-RR (same clone as 82894-7-PBS)

  Various lysates were subjected to SDS PAGE followed by western blot with 82894-7-RR (NSUN2 antibody) at dilution of 1:10000 incubated at room temperature for 1.5 hours. This data was developed using the same antibody clone with 82894-7-PBS in a different storage buffer formulation.

  IHC staining of human stomach cancer using 82894-7-RR (same clone as 82894-7-PBS)

  Immunohistochemical analysis of paraffin-embedded stomach cancer slide using 82894-7-RR (NSUN2 antibody) at dilution of 1:2000 (under 20x lens). Heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0). This data was developed using the same antibody clone with 82894-7-PBS in a different storage buffer formulation.

  IF Staining of HepG2 using 82894-7-RR (same clone as 82894-7-PBS)

  Immunofluorescent analysis of (4% PFA) fixed HepG2 cells using NSUN2 antibody (82894-7-RR, Clone: 230186H8 ) at dilution of 1:250 and CoraLite®488-Conjugated AffiniPure Goat Anti-Rabbit IgG(H+L) (SA00013-2).CL594-Phalloidin (red). This data was developed using the same antibody clone with 82894-7-PBS in a different storage buffer formulation.

  FC experiment of HeLa using 82894-7-RR (same clone as 82894-7-PBS)

  1x10^6 HeLa cells were intracellularly stained with 0.25 ug NSUN2 Recombinant antibody (82894-7-RR, Clone:230186H8) and CoraLite®488-Conjugated AffiniPure Goat Anti-Rabbit IgG(H+L) (SA00013-2)(red), or 0.25 ug Isotype Control (blue). Cells were fixed and permeabilized with Transcription Factor Staining Buffer Kit (PF00011). This data was developed using the same antibody clone with 82894-7-PBS in a different storage buffer formulation.

  Affinity and Kinetic Characterization of 82894-7-RR

  Biolayer interferometry (BLl) kinetic assays of 82894-7-RR against Human NSUN2 were performed. The affinity constant is below 1 pM.

Informations générales

NSUN2, also known as SAKI or Misu (Myc-induced SUN-domain-containing protein), is a methyltransferase which catalyses (cytosine-5-)-methylation of tRNA. NSUN2 is direct target gene of c-Myc and may act downstream of Myc to regulate epidermal cell growth and proliferation. NSUN2 is overexpressed in various cancer tissues and may be a valuable target for cancer therapy and a cancer diagnostic marker. Recently a splicing mutation in NSUN2 has been identified as the cause of a Dubowitz-like syndrome, an autosomal recessive disorder.
