  Non-Neutralization experiment of OCI-AML5 cells using 69534-1-Ig

  Recombinant human IL-27(Cat.NO. HZ-1275) stimulates proliferation of OCI-AML5 cells in a dose-dependent manner (blue curve, refer to bottom X-left Y). The activity of human IL-27 (250 ng/mL) is neutralized by mouse anti-human IL-27 monoclonal antibody 69034-1-Ig at serial dose (red curve, refer to top X-right Y). The ND50 is typically 0.4-1 μg/mL.The control mouse anti-human IL-27 monoclonal antibody 69534-1-Ig could recognize human IL-27, but could not neutralize human IL-27 at serial does at the range of 0.0033-9μg/mL(green curve, refer to top X-right Y).

  ELISA experiment of Recombinant protein using 69534-1-Ig

  Indirect ELISA was carried out by coating recombinant Human IL-27 (Cat.NO. HZ-1275) at 70 ng/well followed by blocking and adding serial diluted IL-27 antibody 69034-1-Ig and 69534-1-Ig respectively. Signal was developed with TMB and stopped by H2SO4. Signal strength was measured by absorbance at 450 nm.

IL-27 is a heterodimeric cytokine composed of two subunits: IL-27a and IL-27b. IL-27 acts on various cell types, including T cells, B cells, macrophages, dendritic cells, natural killer (NK) cells and non-hematopoietic cells. IL-27 plays a critical role in the early regulation of T helper type 1 initiation, and enhances proliferation of naive CD4+T cells and naive B cells. It, however, also exerts anti-inflammatory functions by inhibiting the development of Th17 cells and inducing IL-10 producing type 1 regulatory T cells. IL-27 is a potentially promising cytokine for therapeutic approaches on various human diseases.

This antibody is a neutralizing control antibody for IL-27, the immunogen is the same as Neutrakine 69034-1-Ig but could not neutralize human IL-27.
