  Functional Assay experiment of Recombinant protein using 65684-2-MR

  Inhibition assay of CD279 using 65684-2-MR by ELISA. Mouse PD-1 (Eg0986) was coated at 5 ug/mL. Serial diluted 65684-2-MR and 5 ug/ml biotin-labelled mouse PD-L1 was applied, and the binding signal of PD-L1 to PD-1 was detected by HRP-SA. Signal was developed by TMB substrate. The IC50 is 0.05496 ug/ml.

  FC experiment of mouse splenocytes using 65684-2-MR

  1x10^6 untreated or anti-CD3/CD28 treated mouse splenocytes were surface stained with 0.25 ug Anti-Mouse PD-1/CD279 (RMP1-14) Mouse IgG2a RecAb (65684-2-MR, Clone:RMP1-14) and PE-conjugated Goat Anti-Mouse IgG2a. Cells were then stained with CoraLite® Plus 647 Anti-Mouse CD3. Cells were not fixed.

  1x10^6 anti-CD3/CD28 treated mouse splenocytes were surface stained with 0.25 ug Anti-Mouse PD-1/CD279 (RMP1-14) Mouse IgG2a RecAb (65684-2-MR, Clone: RMP1-14) (red) or Mouse IgG2a Isotype Control (C1.18.4) (65208-1-Ig, Clone: C1.18.4) (blue), and PE-conjugated Goat Anti-Mouse IgG2a. 1x10^6 untreated mouse splenocytes were surface stained with 0.25 ug Anti-Mouse PD-1/CD279 (RMP1-14) Mouse IgG2a RecAb (65684-2-MR, Clone:RMP1-14) (black, dashed) and PE-conjugated Goat Anti-Mouse IgG2a. Cells were not fixed.

  Blocking experiment of Recombinant protein using 65684-2-MR

  Inhibition assay of PD-1(CD279) using 65684-2-MR by Biolayer interferometry (BLI). Biolayer interferometry (BLI) inhibition assay confirms that 65684-2-MR efficiently blocks the interaction between PD-1 and PD-L1. During the assay, mouse PD-1 (Eg0918) was immobilized, incubated with 65684-2-MR to saturation, and then exposed to mouse PD-L1 (Eg0985). The results show an inhibition rate of 0.384, equivalent to 61.6% suppression of PD-L1 binding, demonstrating the strong potential of 65684-2-MR as a PD-1/PD-L1 interaction inhibitor.

  Blocking experiment of Recombinant protein using 65684-2-MR

  Biolayer interferometry (BLl) epitope binding assay of 65684-2-MR and 98080-1-RR. Biolayer interferometry (BLl) epitope binning assay revealed distinct binding epitopes for 65684-2-MR and 98080-1-RR. 5 ug/ml mouse PD-1 (Eg0918) was immoblized onto anti-His probe, followed by binding to 65684-2-MR until saturation, and then the binding signal of 98080-1-RR with binded PD-1 complex was detected. The inhibition rate (the rate of inhibited response to uninhibited response) is 1.026.

  Affinity and Kinetic Characterization of 65684-2-MR

  Biolayer interferometry (BLl) kinetic assays of 65684-2-MR against Mouse PD-1/CD279 were performed. The affinity constant is below 1 pM.

Programmed cell death 1 (PD-1, also known as CD279) is an immunoinhibitory receptor that belongs to the CD28/CTLA-4 subfamily of the Ig superfamily. It is a 288 amino acid (aa) type I transmembrane protein composed of one Ig superfamily domain, a stalk, a transmembrane domain, and an intracellular domain containing an immunoreceptor tyrosine-based inhibitory motif (ITIM) as well as an immunoreceptor tyrosine-based switch motif (ITSM) (PMID: 18173375). PD-1 is expressed during thymic development and is induced in a variety of hematopoietic cells in the periphery by antigen receptor signaling and cytokines (PMID: 20636820). Engagement of PD-1 by its ligands PD-L1 or PD-L2 transduces a signal that inhibits T-cell proliferation, cytokine production, and cytolytic function (PMID: 19426218). It is critical for the regulation of T cell function during immunity and tolerance. Blockade of PD-1 can overcome immune resistance and also has been shown to have antitumor activity (PMID: 22658127; 23169436). RMP1-14 is a monoclonal antibody that targets the murine PD-1 protein and can be used in mouse models to block the reaction between PD-1 and PD-L1/PD-L2 (PMID: 14764726; 31832706).
