  WB analysis using 85087-5-RR

  Various lysates were subjected to SDS PAGE followed by western blot with 85087-5-RR (PDIA3 antibody) at dilution of 1:10000 incubated at room temperature for 1.5 hours.

• 

  IHC staining of human lung cancer using 85087-5-RR

  Immunohistochemical analysis of paraffin-embedded human lung cancer tissue slide using 85087-5-RR (PDIA3 antibody) at dilution of 1:400 (under 20x lens). Heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0).

  FC experiment of HeLa using 85087-5-RR

  1x10^6 HeLa cells were intracellularly stained with 0.25 ug PDIA3 Recombinant antibody (85087-5-RR, Clone:242735H7) and CoraLite®488-Conjugated Goat Anti-Rabbit IgG(H+L) (SA00013-2)(red), or 0.25 ug Rabbit IgG Isotype Control RecAb (98136-1-RR, Clone: 240953C9) (blue). Cells were fixed with 4% PFA and permeabilized with Flow Cytometry Perm Buffer (PF00011-C).

  Affinity and Kinetic Characterization of 85087-5-RR

  Biolayer interferometry (BLl) kinetic assays of 85087-5-RR against Human PDIA3 were performed. The affinity constant is 13.0 nM.

Informations générales

PDIA3, also named as P58, ER60, ERp57, ERp60, ERp61, GRP57, GRP58 and PI-PLC, is a member of the PDI family, It participates in the oxidation, reduction, and isomerization of disulfide bonds for correct folding of secretory proteins before modification and transport in the endoplasmic reticulum. It is associated with apoptosis or inhibition of cancer cell growth. PDIA3 was once thought to be a phospholipase; however, it has been demonstrated that this protein actually has protein disulfide isomerase activity. It is thought that complexes of lectins and PDIA3 mediate protein folding by promoting formation of disulfide bonds in their glycoprotein substrates.
