  WB analysis using 14580-1-AP

  CTLL-2 cells were subjected to SDS PAGE followed by western blot with 14580-1-AP (Perforin antibody) at dilution of 1:10000 incubated at room temperature for 1.5 hours.

• 

  WB analysis of mouse kidney using 14580-1-AP

  mouse kidney tissue were subjected to SDS PAGE followed by western blot with 14580-1-AP (PRF1 antibody) at dilution of 1:500 incubated at room temperature for 1.5 hours.

• 

  FC experiment of NK92 using 14580-1-AP

  1X10^6 NK-92 were intracellularly stained with 0.4 ug Anti-Human Perforin (14580-1-AP) and CoraLite®488-Conjugated AffiniPure Goat Anti-Rabbit IgG(H+L) at dilution 1:1000 (red), or 0.4 ug isotype control. Cells were fixed with 4% PFA and permeabilized with Flow Cytometry Perm Buffer (PF00011-C).

Informations générales

Perforin 1 (PRF1) is one of the major cytolytic proteins of cytolytic granules. It is known to be a crucial effector molecule in cytolytic T lymphocyte and natural killer cell-mediated cytotoxicity. This protein has structural and functional similarities to complement component C9. Like C9, this protein creates transmembrane tubules and is capable of lysing nonspecifically a variety of target cells. Defects in PRF1 are the cause of familial hemophagocytic lymphohistiocytosis type 2 (FHL2), which is characterized by immune dysregulation with hypercytokinemia and defective natural killer cell function.
