  WB analysis using 85238-1-RR

  Non-treated NIH/3T3 cells and TNF alpha (HZ-1014) treated NIH/3T3 cells were subjected to SDS PAGE followed by western blot with 85238-1-RR (Phospho-IRS1 (Ser307) antibody) at dilution of 1:5000 incubated at room temperature for 1.5 hours. The membrane was stripped and re-blotted with Lamin B1 (66095-1-Ig) antibody as a loading control.

Serine phosphorylation of insulin receptor substrate-1 (IRS-1) inhibits insulin signal transduction in a variety of cell backgrounds, which might contribute to peripheral insulin resistance. However, because of the large number of potential phosphorylation sites, the mechanism of inhibition has been difficult to determine. It has been confirmed that JNK and IKK are involved in the phosphorylation of IRS-1 (Ser307/312). Additionally, activation of Akt, ERK, mTOR, and PKCζ correlates to IRS-1 phosphorylation at Ser307/312, Ser267/270, and Ser612/616 in rodent/human IRS-1. (PMID: 11606564, PMID: 12714600)
