  WB analysis using 84147-1-RR (same clone as 84147-1-PBS)

  Non-treated NIH/3T3 cells, Anisomycin treated and UV treated NIH/3T3 cells were subjected to SDS PAGE followed by western blot with 84147-1-RR (Phospho-JUN (Ser63) antibody) at dilution of 1:2000 incubated at room temperature for 1.5 hours. The membrane was stripped and re-blotted with Beta Actin (66009-1-Ig) antibody as a loading control. This data was developed using the same antibody clone with 84147-1-PBS in a different storage buffer formulation.

  ChIP experiment of PC-12 using 84147-1-RR (same clone as 84147-1-PBS)

  Chromatin was prepared from PC-12 cells starved overnight and treated with Human β-Nerve Growth Factor (hβ-NGF) (50 ng/ml, 2 hr). Cells were fixed with formaldehyde for 10 minutes. The ChIP was performed with 20 µg of cross-linked chromatin, 5 µg of Phospho-JUN (Ser63) (84147-1-RR) or 5 ug of Normal Rabbit IgG (98136-1-RR), and 20 µl of Protein A Magarose Beads. The immunoprecipitated DNA was quantified by real-time PCR. This data was developed using the same antibody clone with 84147-1-PBS in a different storage buffer formulation.

JUN is also named as c-Jun and AP1, belongs to the bZIP family and Jun subfamily. JUN, the most extensively studied protein of the activator protein-1 (AP-1) complex, is involved in numerous cell activities, such as proliferation, apoptosis, survival, tumorigenesis and tissue morphogenesis.The phosphorylation of c-Jun by Plk3 immunoprecipitates was not altered by the pre-removal of JNK from the cell lysates. In addition, the effect of UV irradiation-induced phosphorylation of c-Jun and apoptosis were not significantly affected by knockdown of JNK mRNA. (PMID: 22180088, PMID: 17804415)
