  WB analysis of HeLa using 80199-2-RR (same clone as 80199-2-PBS)

  Non-treated HeLa cells, phosphatase inhibitor treated HeLa cells and λ phosphatase treated HeLa cells were subjected to SDS PAGE followed by western blot with 80199-2-RR (Phospho-STAT3 (Ser727) antibody) at dilution of 1:2000 incubated at room temperature for 1.5 hours. The membrane was stripped and reblotted with GAPDH Monoclonal antibody (60004-1-Ig) as loading control. This data was developed using the same antibody clone with 80199-2-PBS in a different storage buffer formulation.

  FC experiment of HEK-293T using 80199-2-RR (same clone as 80199-2-PBS)

  1x10^6 Calyculin A treated HEK-293T cells were intracellularly stained with 0.25 ug Stat3 Recombinant Antibody (80199-2-RR, Clone:240778A12) and PE-Conjugated Goat Anti-Rabbit IgG(H+L)(red), or 0.25 ug Isotype Control (blue). Cells were fixed with 4% PFA and permeabilized with 90% MeOH. This data was developed using the same antibody clone with 80199-2-PBS in a different storage buffer formulation.

  ChIP experiment of HepG2 using 80199-2-RR (same clone as 80199-2-PBS)

  Chromatin was prepared from HepG2 cells (starved overnight) either non-treated or treated with IL-6 (100ng/mL, 30min). Cells were fixed with formaldehyde for 10 minutes. The ChIP was performed with 15 µg of cross-linked chromatin, 5 µg of Phospho-STAT3 (Ser727) (80199-2-RR) or 5 ug of Normal Rabbit IgG (98136-1-RR), and 20 µl of Protein A Magarose Beads. The immunoprecipitated DNA was quantified by real-time PCR. This data was developed using the same antibody clone with 80199-2-PBS in a different storage buffer formulation.

Informations générales

Signal transducer and activator of transcription 3 (acute-phase response factor) (STAT3, synonyms: APRF, FLJ20882, MGC16063) is a member of the STAT protein family. In response to cytokines and growth factors, STAT family members are phosphorylated by the receptor associated kinases, and then form homo- or heterodimers that translocate to the cell nucleus where they act as transcription activators. STAT3 is activated through phosphorylation in response to various cytokines and growth factors including IFNs, EGF, IL5, IL6, HGF, LIF and BMP2. STAT3 mediates the expression of a variety of genes in response to cell stimuli, and thus plays a key role in many cellular processes such as cell growth and apoptosis.
