  WB analysis using 82382-2-RR (same clone as 82382-2-PBS)

  Non-treated and Calyculin A treated HeLa cells were subjected to SDS PAGE followed by western blot with 82382-2-RR (Phospho-TBK1 (Ser172) antibody) at dilution of 1:1000 incubated at room temperature for 1.5 hours. The membrane was stripped and re-blotted with Alpha Tubulin (66031-1-Ig) antibody as a loading control. This data was developed using the same antibody clone with 82382-2-PBS in a different storage buffer formulation.

  FC experiment of HeLa using 82382-2-RR (same clone as 82382-2-PBS)

  1X10^6 HeLa cells untreated (dashed lines) or treated with Calyculin A which intracellularly stained with 0.06 ug Phospho-TBK1 (Ser172) Recombinant antibody (82382-2-RR, Clone:241787G12) and CoraLite®488-Conjugated Goat Anti-Rabbit IgG(H+L) (SA00013-2)(red), or 0.06 ug Rabbit IgG Isotype Control RecAb (98136-1-RR, Clone: 240953C9) (blue). Cells were fixed with 4% PFA and permeabilized with 90% MeOH. This data was developed using the same antibody clone with 82382-2-PBS in a different storage buffer formulation.

Informations générales

The serine/threonine protein kinase TBK1, discovered two decades ago as a member of the IκB kinase (IKK) protein kinase family, plays crucial roles in regulating innate immune response by promoting type I interferon (IFN) and modulating NF-κB signaling. The phosphorylation of Ser172 residue on TBK1 is critical and is commonly used as an indicator for its activity. (PMID: 39279883)
