  FC experiment of human PBMCs using 98139-1-RR (same clone as 98139-1-PBS)

  1x10^6 untreated or IFN-α treated human PBMCs were surface stained with 0.25 ug Anti-Human TRAIL/CD253 Rabbit RecAb (98139-1-RR, Clone: 241419D2) and PE-Conjugated Goat Anti-Rabbit IgG(H+L). Cells were co-stained with CoraLite® Plus 647 Anti-Human CD14 Rabbit Recombinant Antibody (CL647-98040, Clone: 230332D7). Cells were incubated with FC Receptor Block prior to staining. Cells were not fixed. This data was developed using the same antibody clone with 98139-1-PBS in a different storage buffer formulation.

  1x10^6 IFN-α treated human PBMCs were surface stained with 0.25 ug Anti-Human TRAIL/CD253 Rabbit RecAb (98139-1-RR, Clone: 241419D2) (red) or 0.25 ug Rabbit IgG Isotype Control RecAb (98136-1-RR, Clone: 240953C9) (blue), and PE-Conjugated Goat Anti-Rabbit IgG(H+L). 1x10^6 untreated human PBMCs were surface stained with 0.25 ug Anti-Human TRAIL/CD253 Rabbit RecAb (98139-1-RR, Clone: 241419D2) (black, dashed) and PE-Conjugated Goat Anti-Rabbit IgG(H+L). Cells were incubated with FC Receptor Block prior to staining. Cells were not fixed. This data was developed using the same antibody clone with 98139-1-PBS in a different storage buffer formulation.

  Affinity and Kinetic Characterization of 98139-1-RR

  Biolayer interferometry (BLl) kinetic assays of 98139-1-RR against Human TRAIL/CD253 were performed. The affinity constant is below 1 pM.

Informations générales

TRAIL, also known as CD253 or TNFSF10 (tumor necrosis factor superfamily member 10), is a typical death ligand expressed on natural killer cells and cytotoxic T lymphocytes. This protein preferentially induces apoptosis in transformed and tumor cells, but does not appear to kill normal cells although it is expressed at a significant level in most normal tissues. TRAIL induces apoptotic cell death in cancer by binding to its functional death receptors, death receptor (DR) 4 (TNFRSF10A/TRAIL-R1) and DR5 (TNFRSF10B/TRAIL-R2) to activate the extrinsic apoptosis pathway. TRAIL also activates c-Jun N-terminal kinase (MAPK8/JNK) and the transcription factor nuclear factor-κB (NFκB). The binding of this protein to its receptors has been shown to trigger the activation of MAPK8/JNK, caspase 8, and caspase 3.
