  WB analysis using 83240-5-RR (same clone as 83240-5-PBS)

  Various lysates were subjected to SDS PAGE followed by western blot with 83240-5-RR (USP30 antibody) at dilution of 1:10000 incubated at room temperature for 1.5 hours. This data was developed using the same antibody clone with 83240-5-PBS in a different storage buffer formulation.

  WB analysis of HepG2 using 83240-5-RR (same clone as 83240-5-PBS)

  WB result of USP30 antibody (83240-5-RR; 1:3000; incubated at room temperature for 1.5 hours) with sh-Control and sh-USP30 transfected HepG2 cells. This data was developed using the same antibody clone with 83240-5-PBS in a different storage buffer formulation.

  FC experiment of HeLa using 83240-5-RR (same clone as 83240-5-PBS)

  1x10^6 HeLa cells were intracellularly stained with 0.25 ug USP30 Recombinant antibody (83240-5-RR, Clone:240111C6) and CoraLite®488-Conjugated AffiniPure Goat Anti-Rabbit IgG(H+L) (SA00013-2)(red), or 0.25 ug Isotype Control (blue). Cells were fixed with 4% PFA and permeabilized with Flow Cytometry Perm Buffer (PF00011-C). This data was developed using the same antibody clone with 83240-5-PBS in a different storage buffer formulation.

  Affinity and Kinetic Characterization of 83240-5-RR

  Biolayer interferometry (BLl) kinetic assays of 83240-5-RR against Human USP30 were performed. The affinity constant is below 1 pM.

Informations générales

USP30, a mitochondrion-localized deubiquitylase, regulates mitophagy. Mitochondrial damage stimulates parkin to assemble Lys 6, Lys 11 and Lys 63 chains on mitochondria, and that USP30 is a ubiquitin-specific deubiquitylase with a strong preference for cleaving Lys 6- and Lys 11-linked multimers. USP30 can be ubiquitinated by parkin (PARK2) at Lys-235 and Lys-289. USP30 inhibition is potentially beneficial for treating Parkinson disease by promoting mitochondrial clearance and quality control. USP30 was detected 67 kDa by ubiquitination (PMID: 34704599).
