  WB result of MYC antibody (80845-1-RR; 1:600; incubated at room temperature for 1.5 hours) with sh-Control and sh-MYC transfected A549 cells. This data was developed using the same antibody clone with 80845-1-PBS in a different storage buffer formulation.

  1x10^6 HeLa cells were intracellularly stained with 0.25 ug c-MYC Recombinant antibody (80845-1-RR, Clone:2B17) and CoraLite®488-Conjugated Goat Anti-Rabbit IgG(H+L) (SA00013-2)(red), or 0.25 ug Rabbit IgG Isotype Control RecAb (98136-1-RR, Clone: 240953C9) (blue). Cells were fixed and permeabilized with Transcription Factor Staining Buffer Kit (PF00011). This data was developed using the same antibody clone with 80845-1-PBS in a different storage buffer formulation.

  ChIP experiment of HeLa using 80845-1-RR (same clone as 80845-1-PBS)

  Chromatin was prepared from HeLa cells. Cells were fixed with formaldehyde for 10 minutes. The ChIP was performed with 20 µg of cross-linked chromatin, 5 µg of c-MYC (80845-1-RR) or 5 ug of Normal Rabbit IgG (98136-1-RR), and 20 µl of Protein A Magarose Beads. The immunoprecipitated DNA was quantified by real-time PCR. This data was developed using the same antibody clone with 80845-1-PBS in a different storage buffer formulation.

  Affinity and Kinetic Characterization of 80845-1-RR

  Biolayer interferometry (BLl) kinetic assays of 80845-1-RR against Human c-MYC were performed. The affinity constant is 2.03 nM.

Informations générales

MYC contains one basic helix-loop-helix (bHLH) domain. This protein is a multifunctional, nuclear phosphoprotein that plays a role in cell cycle progression, apoptosis and cellular transformation. It functions as a transcription factor that regulates transcription of specific target genes. It seems to activate the transcription of growth-related genes. It binds DNA in a non-specific manner, yet also specifically recognizes the core sequence 5'-CAC[GA]TG-3'. Mutations, overexpression, rearrangement and translocation of this gene have been associated with a variety of hematopoietic tumors, leukemias and lymphomas, including Burkitt lymphoma.
