  FC experiment of MCF-7 using 21903-1-AP

  1X10^6 MCF-7 cells were stained with 0.2ug Nectin-4/PVRL4 antibody (21903-1-AP, red) and control antibody (blue). Fixed with 4% PFA blocked with 3% BSA (30 min). Alexa Fluor 488-conjugated AffiniPure Goat Anti-Rabbit IgG(H+L) with dilution 1:1500.

Informations générales

Nectin-4, or PVRL4, belongs to the family of cell adhesion molecules that regulate the formation of adherence functions in epithelial cells through the AF-6 afadin scaffold molecule. Nectin-4 is reported as a histological and serological marker for breast carcinoma. Nectin-4 can be detected as a soluble form of 43 kDa and a transmembrane form of 66 kDa (PMID: 15784625).

I had a lot of trouble using this PVRL4 antibody on western blots of 2 ovarian cancer cell lines; OV8 and OV90. I kept getting prominent bands at 250 kda. I tried cutting the 250 band to see if that would help, but it did not make the signal any better in the band(s) below. Any insight on this would be greatly appreciated. Thank you.
