  1x10^6 THP-1 cells were intracellularly stained with 0.25 ug Anti-Human EP300 (83078-5-RR, Clone:230369D12) and APC-Conjugated AffiniPure Goat Anti-Rabbit IgG(H+L)(red), or 0.25 ug rabbit IgG isotype control (blue). Cells were fixed and permeabilized with True-Nuclear Transcription Factor Buffer Set. This data was developed using the same antibody clone with 83078-5-PBS in a different storage buffer formulation.

  ChIP experiment of K-562 using 83078-5-RR (same clone as 83078-5-PBS)

  Chromatin was prepared from K-562 cells. Cells were fixed with formaldehyde for 10 minutes. The ChIP was performed with 20 µg of cross-linked chromatin, 5 µg of p300 (83078-5-RR) or 5 ug of Normal Rabbit IgG (98136-1-RR), and 20 µl of Protein A Magarose Beads. The immunoprecipitated DNA was quantified by real-time PCR. This data was developed using the same antibody clone with 83078-5-PBS in a different storage buffer formulation.

Informations générales

p300 is a 300 kDa global transcriptional coactivator with histone acetyltransferase (HAT) activity. p300 binds to chromosomal histones and modulates the chromatin conformation by acetylation of its histones, leading to activation of various gene transcriptions. p300 plays a critical role in DNA repair synthesis after DNA damage induced by ultraviolet or radiation. p300 is overexpressed in various types of cancer, which is associated with worse patient outcomes (PMID: 27322077).
