  FC experiment of human PBMCs using AY7-FcA98211

  1x10^6 untreated or IFN gamma, TNF α and Brefeldin A treated human PBMCs were intracellularly stained with FcZero-rAb™ CoraLite® Plus 488 Anti-Human CD14 and 5 ul APC-Cyanine7 Anti-Human CXCL9/MIG Rabbit RecAb (AY7-FcA98211, Clone:241437F5). Cells were incubated with MonoZero™ Monocytes blocking Reagent(PF00020) prior to staining. Cells were fixed with 4% PFA and permeabilized with Flow Cytometry Perm Buffer (PF00011-C).

  1x10^6 IFN gamma, TNF α and Brefeldin A treated human PBMCs were intracellularly stained with 5 ul APC-Cyanine7 Anti-Human CXCL9/MIG Rabbit RecAb (AY7-FcA98211, Clone: 241437F5) (red) or FcZero-rAb™ APC-Cyanine7 Rabbit IgG Isotype Control Recombinant Antibody (AY7-FcA98136, Clone: 240953C9) (blue). 1x10^6 untreated human PBMCs were intracellularly stained with 5 ul APC-Cyanine7 Anti-Human CXCL9/MIG Rabbit RecAb (AY7-FcA98211, Clone: 241437F5) (black, dashed). Cells were incubated with MonoZero™ Monocytes blocking Reagent(PF00020) prior to staining. Cells were fixed with 4% PFA and permeabilized with Flow Cytometry Perm Buffer (PF00011-C).

Hintergrundinformationen

CXCL9 is a small cytokine known as MIG. CXCL9 is a T-cell chemoattractant, which is induced by IFN-γ. It is closely related to two other CXC chemokines called CXCL10 and CXCL11, whose genes are located near the gene for CXCL9 on human chromosome 4. A high level of CXCL9 revealed in peripheral liquids, can be considered as a marker of host immune response, especially of that involving Th1 cells.
