  1x10^6 Transfected HEK-293 cells were intracellularly stained with 0.25 ug HA Tag Recombinant antibody (81290-1-RR, Clone:6I21) and CoraLite®488-Conjugated Goat Anti-Rabbit IgG(H+L) (SA00013-2), and 0.25 ug Rabbit IgG Isotype Control RecAb (98136-1-RR, Clone: 240953C9). Cells were fixed with 4% PFA and permeabilized with Flow Cytometry Perm Buffer (PF00011-C).

  Affinity and Kinetic Characterization of 81290-1-RR

  Biolayer interferometry (BLl) kinetic assays of 81290-1-RR against a protein with HA tag were performed. The affinity constant is 0.766 nM.

The HA tag corresponds to amino acid residues YPYDVPDYA of a surface glycoprotein -human influenza virus hemagglutinin (HA). The HA tag is commonly used for a variety of research applications including chromatin immunoprecipitation, ELISA, flow cytometry, western blotting, immunocytochemistry/immunofluorescence among of others.

Did not give a reliable signal in our positive cell line (CRISPRi that should have HA-tagged dCas9-KRAB). Additionally, there is quite some background as in both the positive and the negative line (WTC11), the nuclei are stained.

This HA Tag Recombinant antibody (81290-1-RR) is by far the best HA antibody I have used. It worked exceptionally well in my hands for immunofluorescence experiments. I tested it on a stable cell line expressing an HA-tagged protein and on cells transiently transfected with an HA-tagged protein, and the results were consistently outstanding. The staining was crisp and specific, with minimal background. I used it at a 1:500 dilution on U2OS cells and achieved excellent results.
