  WB analysis using 84876-5-RR

  Various lysates were subjected to SDS PAGE followed by western blot with 84876-5-RR (NNMT antibody) at dilution of 1:5000 incubated at room temperature for 1.5 hours.

  1x10^6 HepG2 cells were intracellularly stained with 0.25 ug NNMT Recombinant antibody (84876-5-RR, Clone:242394B7) and APC-Conjugated Goat Anti-Rabbit IgG(H+L)(red), or 0.25 ug Rabbit IgG Isotype Control Recombinant Antibody (98136-1-RR, Clone: 240953C9) (blue). Cells were fixed with 4% PFA and permeabilized with Flow Cytometry Perm Buffer (PF00011-C).

  Affinity and Kinetic Characterization of 84876-5-RR

  Biolayer interferometry (BLl) kinetic assays of 84876-5-RR against Human NNMT were performed. The affinity constant is 0.824 nM.

NNMT can catalyze the N-methylation of nicotinamide using the universal methyl donor S-adenosyl-L-methionine to form N1-methylnicotinamide and S-adenosyl-L-homocysteine, a predominant nicotinamide/vitamin B3 clearance pathway (PMID: 21823666; 23455543; 8182091). It plays a central role in regulating cellular methylation potential, by consuming S-adenosyl-L-methionine and limiting its availability for other methyltransferases. Actively mediates genome-wide epigenetic and transcriptional changes through hypomethylation of repressive chromatin marks, such as H3K27me3 (PMID: 23455543; PMID: 26571212; PMID: 31043742).
