  WB analysis using 83027-4-RR

  Various lysates were subjected to SDS PAGE followed by western blot with 83027-4-RR (SCN4B antibody) at dilution of 1:2000 incubated at room temperature for 1.5 hours.

• 

  IHC staining of rat dorsal root ganglion using 83027-4-RR

  Immunohistochemical analysis of paraffin-embedded rat dorsal root ganglion tissue slide using 83027-4-RR (SCN4B antibody) at dilution of 1:1000 (under 40x lens). Heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0).

• 

  FC experiment of HeLa using 83027-4-RR

  1x10^6 HeLa cells were intracellularly stained with 0.25 ug Anti-Human SCN4B (83027-4-RR, Clone:230297B11) and APC-Conjugated AffiniPure Goat Anti-Rabbit IgG(H+L)(red), or 0.25 ug rabbit IgG isotype control (blue). Cells were fixed with 4% PFA and permeabilized with Flow Cytometry Perm Buffer (PF00011-C).

Mammalian sodium channels are heterotrimers, composed of a central, pore-forming alpha subunit and two auxiliary beta subunits (PMID: 11486343). SCN4B (sodium channel subunit beta-4) is one of several sodium channel beta subunits, which modulate channel gating kinetics. SCN4B has an N-terminal signal peptide, a large extracellular domain predicted to form an Ig-like fold, a single transmembrane alpha helix, and a short cytoplasmic C terminus. Defects in SCN4B gene are a cause of long QT syndrome type 10 (LQT10).
