  WB analysis using 14465-1-AP

  Various lysates were subjected to SDS PAGE followed by western blot with 14465-1-AP (SMC6 antibody) at dilution of 1:6000 incubated at room temperature for 1.5 hours.

• 

  WB analysis of mouse testis using 14465-1-AP

  mouse testis tissue were subjected to SDS PAGE followed by western blot with 14465-1-AP (SMC6 antibody) at dilution of 1:2000 incubated at room temperature for 1.5 hours.

• 

  IP experiment of mouse testis using 14465-1-AP

  IP result of anti-SMC6 (IP:14465-1-AP, 4ug; Detection:14465-1-AP 1:1000) with mouse testis tissue lysate 4000ug.

  Immunohistochemical analysis of paraffin-embedded mouse testis tissue slide using 14465-1-AP (SMC6 antibody) at dilution of 1:200 (under 40x lens). Heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0).

Empfohlene Verdünnung

• Immunpräzipitation (IP): IP : 0.5-4.0 ug for 1.0-3.0 mg of total protein lysate
• Immunhistochemie (IHC): IHC : 1:50-1:500

It is recommended that this reagent should be titrated in each testing system to obtain optimal results.

Hintergrundinformationen

SMC6 (Structural maintenance of chromosomes protein 6) is core component of the SMC5-SMC6 complex that is involved in DNA double-strand breaks by homologous recombination. The complex may promote sister chromatid homologous recombination by recruiting the SMC1-SMC3 cohesin complex to double-strand breaks. The complex is required for telomere maintenance via recombination in ALT (alternative lengthening of telomeres) cell lines and mediates sumoylation of shelterin complex (telosome) components which is proposed to lead to shelterin complex disassembly in ALT-associated PML bodies (APBs) (PMID: 26983541).
