Background Information
SP6 (also known as Epfn) is a C2H2-type zinc-finger transcription factor located at 17q21.32, which functions as a transcriptional activator and plays essential roles in odontogenesis and cell proliferation. Notably, this gene has also been referred to as KLF14 in earlier literature, particularly in the context of its discovery as a member of the SP/XKLF transcription factor family. However, it is critical to distinguish this 17q21.32 SP6/KLF14 from the distinct KLF14 gene located at 7q32.2 (official symbol KLF14, also known as BTEB5), which is an intronless gene encoding a transcriptional co-repressor that exhibits maternal imprinting and is primarily associated with metabolic regulation, including type 2 diabetes and lipid homeostasis, as well as roles in cancer and fibrosis. Despite sharing the "KLF14" alias and structural homology within their zinc-finger DNA-binding domains, these two genes reside at separate chromosomal loci, possess divergent transcriptional activities (activator versus repressor), and govern distinct physiological processes.
