  FC experiment of human PBMCs using 98363-1-RR

  1x10^6 untreated or PMA, Ionomycin and Brefeldin A treated human PBMCs were intracellularly stained with 0.25 ug Anti-Human IL-3 Rabbit RecAb (98363-1-RR, Clone: 240417E1) and PE-Conjugated Goat Anti-Rabbit IgG(H+L). Cells were co-stained with CoraLite® Plus 647 Anti-Human CD4. Cells were fixed with 4% PFA and permeabilized with Flow Cytometry Perm Buffer (PF00011-C). Treated human PBMCs were pre-stimulated with anti-human CD3/CD28 and recombinant human IL-2 and IL-4.

  1x10^6 PMA, Ionomycin and Brefeldin A treated human PBMCs were intracellularly stained with 0.25 ug Anti-Human IL-3 Rabbit RecAb (98363-1-RR, Clone: 240417E1) (red) or Rabbit IgG Isotype Control RecAb (98136-1-RR, Clone: 240953C9) (blue), and PE-Conjugated Goat Anti-Rabbit IgG(H+L). Untreated human PBMCs were intracellularly stained with 0.25 ug Anti-Human IL-3 Rabbit RecAb (98363-1-RR, Clone: 240417E1) and PE-Conjugated Goat Anti-Rabbit IgG(H+L) (black, dashed). Cells were fixed with 4% PFA and permeabilized with Flow Cytometry Perm Buffer (PF00011-C). Treated human PBMCs were pre-stimulated with anti-human CD3/CD28 and recombinant human IL-2 and IL-4.

Interleukin-3 (IL-3) is a multilineage hematopoietic growth factor that promotes the proliferation, differentiation and survival of early multilineage hematopoietic progenitors. In particular, this cytokine plays a key role in stimulating the proliferation and survival of myeloid precursors. It is involved in a variety of cell activities such as cell growth, differentiation and apoptosis. This cytokine has been shown to also possess neurotrophic activity, and it may be associated with neurologic disorders. (PMID: 24103757;2698876;2809196)
