Informations générales

Vinculin belongs to the vinculin/alpha-catenin family. It is an actin filament (F-actin)-binding protein which involved in cell-matrix adhesion and cell-cell adhesion. Vinculin regulates cell-surface E-cadherin expression and potentiates mechanosensing by the E-cadherin complex. It may also play important roles in cell morphology and locomotion. Vinculin is a 117-kDa, 1,066-amino-acid protein which is ubiquitously expressed. Its splice variant, metavinculin (124 kDa), is muscle-specific.

Human airway smooth muscle cells fixed after 1 h of plating. Cells were fixed with 4% PFA and permeabilized with 0.1% Triton X 100. Blocked with 0.1% BSA and incubated with primary antibody overnight at 4C at 1:100 dilution. Following day cells were washed and incubated for 1 h with Alexa 594 at 1:400 dilution.

The antibody was used for immunofluorescence staining of 2D cultured cells fixed with 1%PFA and permeabilized with Triton X100. Cells were incubated overnight at +4 degrees celcius. The staining was visible in both epifluorescence microscope and confocal fluorescence microscope. Also, used for immunostaining of formalin-fixed paraffin-embedded (FFPE) skin of mouse. The antigen retrieval was citrate buffer. Primary antibody was incubated overnight at +4 degrees celcius.
