Product Information

KE00577 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human GPX4 ELISA kit is to be used to detect and quantify protein levels of endogenous human GPX4. The assay recognizes human GPX4. An antibody specific for human GPX4 has been pre-coated onto the microwells. The human GPX4 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human GPX4 is added to detect the captured human GPX4 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information

GPX4, originally known as PHGPX (phospholipid hydroperoxide glutathione peroxidase), was initially purified in porcine liver or heart in 1982 as a peroxidation-inhibiting protein on liposomes and biological membranes in the presence of glutathione (GSH). GPX4 can be expressed in a variety of tissues, with the highest content in the testis, affecting the development and function of sperm. The main function of GPX4 is to use GSH as a co-factor to resist lipid peroxidation, thereby protecting the integrity of the membrane.
