Product Information

KE00630 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human INSR ELISA kit is to be used to detect and quantify protein levels of endogenous human INSR. The assay recognizes human INSR. An antibody specific for human INSR has been pre-coated onto the microwells. The human INSR protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human INSR is added to detect the captured human INSR protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information

INSR (Insulin Receptor) is a transmembrane receptor tyrosine kinase, composed of two α subunits and two β subunits linked by disulfide bonds. The α subunits are located extracellularly and contain the insulin-binding sites, while the β subunits span the cell membrane and contain the intracellular tyrosine kinase domains. INSR is a critical protein involved in regulating glucose homeostasis and metabolic processes. The binding of INS to the receptor's extracellular domain results in intracellular activation of a tyrosine-specific kinase activity and the generation of signals that determine the cellular response. In addition to the widely active Sp1 factor, tissue-specific transcription factors have also been implicated in the regulation of the INSR promoter.
