KE00347 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human PD-L2/B7-DC ELISA kit is to be used to detect and quantify protein levels of endogenous human PD-L2/B7-DC. The assay recognizes human PD-L2/B7-DC. An antibody specific for human PD-L2/B7-DC has been pre-coated onto the microwells. The human PD-L2/B7-DC protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human PD-L2/B7-DC is added to detect the captured human PD-L2/B7-DC protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
Programmed cell death ligand 2 (PD-L2, also known as CD273, or B7-DC), is one of the B7 family molecules which are type I transmembrane proteins belonging to the immunoglobulin superfamily. PD-L2 was identified both by its homology to PD-L1 and by analysis of genes differentially expressed in libraries generated from dendritic cells (DC) and activated macrophages. PD-L2 is a second ligand for PD-1, a costimulatory molecule that plays an inhibitory role in regulating T-cell activation. Overexpression of PD-L2 has been reported in malignant tumors and significantly contributes to tumorigenesis and cancer progression.
