KE00464 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human iNOS ELISA kit is to be used to detect and quantify protein levels of endogenous human iNOS. The assay recognizes human iNOS. An antibody specific for human iNOS has been pre-coated onto the microwells. The human iNOS protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human iNOS is added to detect the captured human iNOS protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
Nitric oxide (NO), which is synthesized by nitric oxide synthase (NOS), is the smallest known bioactive molecule, and can be produced by a variety of cells. NO plays an important role in neurotransmission, vascular function, host defense, and immune regulation. Three NOS isoforms have been identified: neuronal nitric oxide synthase (nNOS), inducible nitric oxide synthase (iNOS), and endothelial nitric oxide synthase (eNOS). iNOS is inducible in many types of cells including epithelial, mesenchymal, and myeloid cells. Induction of iNOS expression varies depending on cell type and species. Aberrant expression of iNOS has been documented in different human tumors including head and neck, breast, colon, stomach, and lung cancer. Increased iNOS expression correlates with tumor grade and angiogenesis in breast cancer patients .
