KE10069 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The Mouse sVEGFR1/Flt-1 ELISA kit is to be used to detect and quantify protein levels of endogenous mouse sVEGFR1/Flt-1. The assay recognizes mouse sVEGFR1/Flt-1. An antibody specific for mouse sVEGFR1/Flt-1 has been pre-coated onto the microwells. The mouse sVEGFR1/Flt-1 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another antibody of biotinylated specific for mouse sVEGFR1/Flt-1 is added to detect the captured mouse sVEGFR1/Flt-1 protein. For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information

Vascular endothelial growth factor receptor-1 (VEGFR-1, FLT-1) is a receptor tyrosine kinase belonging to the VEGFR family. VEGF is a key regulator of physiological angiogenesis and has also been implicated in pathological angiogenesis associated with tumors, intraocular neovascular disorders and other conditions. The biological effects of VEGF are mediated by VEGFR-1 and VEGFR-2. Both the two receptors have seven immunoglobulin-like repeats in the extracellular domain, a single transmembrane region and a tyrosine kinase domain. VEGFR-1 binds VEGFA, PIGF and VEGFB, and plays an essential role in the development of embryonic vasculature, the regulation of angiogenesis, cell survival, cell migration, macrophage function, chemotaxis, and cancer cell invasion. Besides the membrane-bound form, VEGFR-1 can also exist as a soluble form (sVEGFR-1) generated by alternative splicing of VEGFR-1 mRNA.
