• 

  WB analysis of A549 using 20000-1-AP

  A549 cells were subjected to SDS PAGE followed by western blot with 20000-1-AP (TGFBR3-Specific antibody) at dilution of 1:300 incubated at room temperature for 1.5 hours.

• 

  WB analysis of A431 using 20000-1-AP

  A431 cells were subjected to SDS PAGE followed by western blot with 20000-1-AP (TGFBR3-Specific antibody) at dilution of 1:300 incubated at room temperature for 1.5 hours.

• 

  FC experiment of A549 using 20000-1-AP

  1X10^6 A549 cells were intracellularly stained with 0.4 ug Anti-Human TGFBR3-Specific (20000-1-AP) and CoraLite®488-Conjugated AffiniPure Goat Anti-Rabbit IgG(H+L) at dilution 1:1000 (red), or 0.4 ug Rabbit IgG control Rabbit PolyAb (30000-0-AP, Clone: ) (blue). Cells were fixed with 4% PFA and permeabilized with Flow Cytometry Perm Buffer (PF00011-C).

Background Information

Transforming growth factor β (TGF-β) is a multifunctional cytokine that plays important roles in normal development and homeostasis (PMID: 8530052). There are three TGF-β forms: TGF-β1, TGF-β2, and TGF-β3. The diverse effects of TGF-β are mediated by the TGF-β receptors and cell surface-binding proteins. Three TGF-beta receptors exist: TGFBR1, TFGBR2, TGFBR3 (PMID: 8530052; 30184463). TGFBR3, also known as betaglycan, is a transmembrane proteoglycan that often functions as a co-receptor with other TGF-beta receptor superfamily members. In addition to the transmembrane form, the extracellular domain of TGFBR3 can be cleaved to a soluble form (sTGFBR3), which may serve as an antagonist of TGF-β to inhibit TGF-β signaling (PMID: 30184463). TGFBR3 has a larger apparent molecular weight than the calculated molecular weight, due to extensive carbohydrate modifications (PMID: 1556106). This antibody is specific to TGFBR3.

This antibody worked well for labeling TGF beta receptor type 3 in mouse kidney. I was able to acquire robust immunofluorescence signals with the antibody and highly recommend this KO validated antibody.
