Product Information

KE00254 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The Human ACE2 ELISA kit is to be used to detect and quantify protein levels of endogenous human ACE2. The assay recognizes human ACE2. An antibody specific for human ACE2 has been pre-coated onto the microwells. The human ACE2 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another antibody for human ACE2 is added to detect the captured human ACE2 protein. For signal development, horseradish peroxidase (HRP)-conjugated antibody is added, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information

Angiotensin-converting enzyme 2 (ACE2), a critical regulator of the renin-angiotensin system (RAS), belongs to the peptidase M2 family. It plays an important role in cardiovascular homeostasis by regulating vascular tone, fluid, and electrolyte balance. ACE2 functions as a carboxymonopeptidase hydrolyzing the cleavage of a single C-terminal residue from Angiotensin-II (Ang-II), the key peptide hormone of RAS, to form Angiotensin-(1-7) (Ang-(1-7)), which binds to the G-protein-coupled Mas receptor and activates signaling pathways that counteract the pathways activated by Ang-II. ACE2 is expressed in a variety of tissues, including the kidneys, testes, heart, and intestines, and is particularly enriched in lung epithelium. Recent studies have shown that increased ACE2 expression is likely to help prevent secondary fibrosis changes following COVID-19 pneumonia.
