KE00359 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human CD28 ELISA kit is to be used to detect and quantify protein levels of endogenous human CD28. The assay recognizes human CD28. An antibody specific for human CD28 has been pre-coated onto the microwells. The human CD28 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human CD28 is added to detect the captured human CD28 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
CD28 (T-cell-specific surface glycoprotein CD28), or T44 and Tp44, is a member of the immunoglobulin superfamily and is expressed on most T lineage cells, NK cell subsets, and plasma cells. CD28 may affect in vivo immune responses as a cell adhesion molecule linking B and T lymphocytes and as the surface component of a novel signal transduction pathway. CD28 binds both CD80 and CD86 with a highly conserved motif MYPPY in the CDR3-like loop. In addition, CD28 is considered a major co-stimulatory molecule, inducing T lymphocyte activation and IL-2 synthesis, and preventing cell death.
