KE00535 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human CCR5 ELISA kit is to be used to detect and quantify protein levels of endogenous human CCR5. The assay recognizes human CCR5. An antibody specific for human CCR5 has been pre-coated onto the microwells. The human CCR5 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human CCR5 is added to detect the captured human CCR5 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
CCR5 (CC chemokine receptor 5) is a seven-transmembrane, G protein-coupled receptor (GPCR) that regulates trafficking and effector functions of memory/effector T-lymphocytes, macrophages, and immature dendritic cells. It also serves as the main coreceptor for the entry of R5 strains of human immunodeficiency virus (HIV-1, HIV-2). CCR5 plays a pivotal role when expressed in breast cancers in facilitating tumor progression and metastasis in multiple ways. CCR5 is also among the most studied chemokine receptors due to its profound significance in human health and disease.
