KE01035 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The Cyclin B1 ELISA kit is to be used to detect and quantify protein levels of endogenous Cyclin B1. The assay recognizes human Cyclin B1. An antibody specific for Cyclin B1 has been pre-coated onto the microwells. The Cyclin B1 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for Cyclin B1 is added to detect the captured Cyclin B1 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
Cyclin B1 belongs to the Cyclin family. Cyclin B1 first appears in S-phase, then accumulates during G2 and mitosis, and finally is degraded rapidly at the onset of anaphase. Cyclin B1 binds CDK1 with high affinity and forms the primary CDK complex necessary for progression and completion of mitosis. Cyclin B1 localizes to kinetochore during prometaphase and contributes to kinetochore-microtubule attachment and efficient alignment of chromosomes. Cyclin B1 could be a mitotic activator of CDK5 that regulates chromosome alignment.
