Product Information

KE01065 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human Frataxin ELISA kit is to be used to detect and quantify protein levels of endogenous human Frataxin. The assay recognizes human Frataxin. An antibody specific for human Frataxin has been pre-coated onto the microwells. The human Frataxin protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human Frataxin is added to detect the captured human Frataxin protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information

Frataxin is a highly conserved iron-binding protein present in most organisms. Frataxin is primarily located in the mitochondria. It is vital for creating iron-sulfur clusters, which are essential for many cellular processes, including the Krebs cycle, electron transport chain components of complexes I to III, and fatty acid β‐oxidation. Reduced levels of the protein frataxin cause the neurodegenerative disease Friedreich's ataxia. Pathology is associated with disruption of iron-sulfur cluster biosynthesis, mitochondrial iron overload, and oxidative stress.
