KE00663 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human IDS ELISA kit is to be used to detect and quantify protein levels of endogenous human IDS. The assay recognizes human IDS. An antibody specific for human IDS has been pre-coated onto the microwells. The human IDS protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human IDS is added to detect the captured human IDS protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
Iduronate 2-sulfatase (IDS) is required for the lysosomal degradation of heparan sulfate and dermatan sulfate. Mutations causing IDS deficiency in humans result in the lysosomal storage of these glycosaminoglycans and Hunter syndrome, an X chromosome-linked disease. IDS is synthesized as two precursor forms of 76 and 90 kDa that are converted, through a 62 kDa intermediate, to 55 and 45 kDa mature polypeptides due to an internal proteolytic cleavage.
