Background Information

Kininogen 1 (also known as high molecular weight kininogen, HMWK, or Fitzgerald factor) is a multidomain glycoprotein found in the blood of most vertebrate. Kininogen 1 is a key component of the kallikrein-kinin system (KKS), which, together with coagulation factor XI, forms the contact activation system of blood coagulation (CAS), also known as the intrinsic pathway of coagulation. This system contributes to thrombin generation primarily under pathologic conditions and can operate independently from the tissue factor (TF) extrinsic pathway. Kininogen 1 and its fragments have been found to have various biological functions. For example, kininogen 1 can regulate leukocyte recruitment, and its domain 5 (kininostatin) can down-regulate endothelial cell proliferation and migration, inhibiting angiogenesis. Additionally, kininogen 1 has been identified as a potential biomarker for early detection of advanced colorectal adenoma and colorectal cancer.
