KE01079 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human PAPP A2 ELISA kit is to be used to detect and quantify protein levels of endogenous human PAPP A2. The assay recognizes human PAPP A2. An antibody specific for human PAPP A2 has been pre-coated onto the microwells. The human PAPP A2 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human PAPP A2 is added to detect the captured human PAPP A2 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
PAPP A2 (Pregnancy-associated Plasma Protein A2) is a metalloproteinase member of the PAPP A protein family and is mainly secreted by the syncytiotrophoblast cells of the placenta. Unlike PAPP A in the same family that regulates the biological activity of insulin-like growth factors (IGFs), PAPP A2 can specifically cleave and inactivate the IGF-binding protein 5 (IGFBP-5), thereby releasing more free IGFs to promote fetal development. Moreover, its concentration significantly increases in the maternal blood during pregnancy, making it a potential new biomarker for assessing placental function and fetal growth restriction.
