KE00981 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human PTH ELISA kit is to be used to detect and quantify protein levels of endogenous human PTH. The assay recognizes human PTH. An antibody specific for human PTH has been pre-coated onto the microwells. The human PTH protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human PTH is added to detect the captured human PTH protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
Parathyroid hormone (PTH) is the main biologically active hormone produced by the parathyroid cells. PTH plays a crucial role in the maintenance of calcium homeostasis directly acting on bone and kidneys and indirectly on the intestine. PTH is best known for its role as a regulator of skeletal remodelling and calcium homeostasis. Recent preclinical studies have documented an effect of PTH to stimulate lipolysis in both adipocytes and liver cells and to cause browning of adipocytes.
