KE00661 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human Siglec-7/CD328 ELISA kit is to be used to detect and quantify protein levels of endogenous human Siglec-7/CD328. The assay recognizes human Siglec-7/CD328. An antibody specific for human Siglec-7/CD328 has been pre-coated onto the microwells. The human Siglec-7/CD328 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human Siglec-7/CD328 is added to detect the captured human Siglec-7/CD328 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
Sialic acid-binding Ig-like lectin 7 (Siglec-7, also known as CD328) is an inhibitory receptor expressed on natural killer (NK) cells, monocytes, macrophages, dendritic cells, and mast cells. It plays a crucial role in regulating immune responses by binding to sialic acid-containing glycans on the surface of target cells, including cancer cells and pathogens. Moreover, Siglec-7 is emerging as a very important regulator of the immune response, which shows that increased expression of sialylated ligands on tumour cells inhibits NK cell activation through engagement of Siglec-7.
