KE00525 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human TCF1/TCF7 ELISA kit is to be used to detect and quantify protein levels of endogenous human TCF1/TCF7. The assay recognizes human TCF1/TCF7. An antibody specific for human TCF1/TCF7 has been pre-coated onto the microwells. The human TCF1/TCF7 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human TCF1/TCF7 is added to detect the captured human TCF1/TCF7 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
TCF7 (Transcription Factor 7), also known as TCF-1 or T-Cell Factor 1, is a member of the T-cell factor/lymphoid enhancer-binding factor (TCF/LEF) family of high mobility group (HMG) box transcriptional activators. TCF7 is part of the Wnt signaling pathway and plays an important role in the differentiation and self-renewal of memory T cells. It has been found that TCF7 is necessary to revive T cells in response to PD-1 blockade against viral infection or cancer. TCF7 is mainly expressed in T-cells and is also detected in proliferating intestinal epithelial cells and in the basal epithelial cells of mammary gland epithelium.
