Background
HER2/ErbB2 is a critical receptor tyrosine kinase protein belonging to the epidermal growth factor receptor (EGFR) family, which also includes HER1 (EGFR), HER3, and HER4. Unlike other members of the EGFR family, HER2/ErbB2 does not have a known ligand and is often referred to as an "orphan receptor." However, it can form homodimers or heterodimers with other EGFR family members, such as HER1, HER3, and HER4, significantly enhancing its signaling potential. This receptor is involved in the regulation of multiple downstream signaling pathways, including the PI3K-Akt and MAPK pathways, which promote cell survival, proliferation, and migration.
