Background Information

PLK4 (Polo-like kinase 4) is also named SAK, STK18 and belongs to the Ser/Thr protein kinase family. It plays a central role in centriole duplication and is a kinase implicated in tumor development, which is an upstream regulator of canonical biogenesis necessary for centriole formation. It has 3 isoforms produced by alternative splicing.

RPE1 WT cells were starved for 2 days. Fixed with -20°C Ethanol for 10 min at room temperature (RT), permeablized with 0.3% PBST(triton), blocked with 1%BSA in PBS for 1h at RT, PLK4 (rb, 1:50) and ARL13B (ms, 1:200) in 1% BSA in PBS diluted, prim AK Incubation for 1h at RT. Sek Ak goat anti rb (green) and goat anti mouse (pink). PLK4 can be found at the Basal Body of cilia and also stains cytoskeleton. PLK4 antibody could be tried with 1:200 dilution, to reduce background.
