Background Information
Phosphatidylinositol 3,4,5-trisphosphate-dependent Rac exchanger 1 protein (PREX1) is a guanine nucleotide exchange factor (GEF) that specifically activates Rac GTPases, particularly Rac1 (PMID: 16243035). PREX1 is activated downstream of G protein-coupled receptors (GPCRs) and receptor tyrosine kinases (RTKs), such as ErbB receptors, and is involved in PI3K/AKT and MAPK signaling pathways (PMID: 28710285; 27358402). It contributes to glucose uptake in adipocytes and promotes tumor cell proliferation, migration, and metastasis in various cancers, including breast, prostate, and melanoma (PMID: 22109529).
