  WB analysis using 28961-1-AP (same clone as 28961-1-PBS)

  Non-treated HT-29 and etoposide treated HT-29 cells were subjected to SDS PAGE followed by western blot with 28961-1-AP (Phospho-P53(Ser15) antibody) at dilution of 1:2000 incubated at 4℃ overnight. The membrane was stripped and re-blotted with GAPDH antibody as loading control. This data was developed using the same antibody clone with 28961-1-PBS in a different storage buffer formulation.

  WB analysis using 28961-1-AP (same clone as 28961-1-PBS)

  Non-treated HSC-T6 cells and UV treated HSC-T6 cells were subjected to SDS PAGE followed by western blot with 28961-1-AP (Phospho-P53 (Ser15) antibody) at dilution of 1:3000 incubated at room temperature for 1.5 hours. This data was developed using the same antibody clone with 28961-1-PBS in a different storage buffer formulation.

  IF Staining of HT-29 using 28961-1-AP (same clone as 28961-1-PBS)

  Immunofluorescent analysis of (4% PFA) fixed non-treated HT-29 and etoposide treated HT-29 cells using Phospho-P53 (Ser15) antibody (28961-1-AP) at dilution of 1:100 and CoraLite®488-Conjugated AffiniPure Goat Anti-Rabbit IgG(H+L). This data was developed using the same antibody clone with 28961-1-PBS in a different storage buffer formulation.

  IF Staining of HT-29 using 28961-1-AP (same clone as 28961-1-PBS)

  Immunofluorescent analysis of (4% PFA) fixed etoposide treated HT-29 cells using Phospho-P53 (Ser15) antibody (28961-1-AP) at dilution of 1:400 and CoraLite®488-Conjugated AffiniPure Goat Anti-Rabbit IgG(H+L), CL594-phalloidin (red). This data was developed using the same antibody clone with 28961-1-PBS in a different storage buffer formulation.

Background Information

The p53 tumor suppressor protein plays a major role in cellular response to DNA damage and other genomic aberrations. Activation of p53 can lead to either cell cycle arrest and DNA repair or apoptosis . DNA damage induces phosphorylation of p53 at Ser15 and Ser20 and leads to a reduced interaction between p53 and its negative regulator, the oncoprotein MDM2. p53 can be phosphorylated by ATM, ATR, and DNA-PK at Ser15 and Ser37. Phosphorylation impairs the ability of MDM2 to bind p53, promoting both the accumulation and activation of p53 in response to DNA damage.
