Product Information
KE20041 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The rat CD4 ELISA kit is to be used to detect and quantify protein levels of endogenous rat CD4. The assay recognizes rat CD4. An antibody specific for rat CD4 has been pre-coated onto the microwells. The rat CD4 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for rat CD4 is added to detect the captured rat CD4 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
CD4 is a transmembrane glycoprotein of the immunoglobulin superfamily (IgSF), containing four extracellular Ig-like domains, a hydrophobic transmembrane region, and a 40-residue cytoplasmic tail. CD4 is expressed in T helper cells, the majority of thymocytes, monocytes, macrophages, and dendritic cells. CD4 is an accessory protein for MHC class-II antigen/T-cell receptor interaction. It is involved in T cell differentiation, development, and activation by interacting with antigen-presenting cells (APCs). The cytoplasmic tail of CD4 is associated with the Lck kinase, which in turn activates the signaling components of the T cell receptor (TCR)-CD3 complexes.
