Background Information
HPSE2 (heparanase 2) is a homolog of HPSE (heparanase 1). HPSE is an endo-β-glucuronidase that cleaves heparan sulfate proteoglycans (HSPGs), which are important components of the extracellular matrix and basement membranes. HPSE2, however, lacks intrinsic heparan sulfate-degrading activity. Instead, it binds to heparan sulfate with higher affinity than HPSE, thereby competing with HPSE for heparan sulfate binding and inhibiting HPSE's enzymatic activity. This competitive inhibition makes HPSE2 a crucial regulator of heparan sulfate metabolism.
