Background Information
TNFRSF14, also known as HVEM (herpesvirus entry mediator), is a type I transmembrane receptor of the tumor necrosis factor receptor (TNFR) superfamily that functions as a key regulator of immune activation and immune checkpoint signaling. It is broadly expressed on T cells, B cells, NK cells, dendritic cells, and epithelial cells. TNFRSF14 interacts with multiple ligands, including LIGHT (TNFSF14), BTLA, and CD160, enabling it to mediate both co-stimulatory and co-inhibitory immune signals depending on the cellular context. Through these bidirectional interactions, HVEM regulates T-cell activation, cytokine production, mucosal immunity, and immune homeostasis. Dysregulation of TNFRSF14 has been implicated in autoimmune disease, chronic inflammation, viral infection, and tumor immune evasion. In the tumor microenvironment, the HVEM-BTLA axis is increasingly recognized as an immune checkpoint pathway that suppresses antitumor immunity, making TNFRSF14 a promising target in cancer immunotherapy and immune modulation research.
