KE00756 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human SP100 ELISA kit is to be used to detect and quantify protein levels of endogenous human SP100. The assay recognizes human SP100. An antibody specific for human SP100 has been pre-coated onto the microwells. The human SP100 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human SP100 is added to detect the captured human SP100 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
SP100 is a protein that performs multiple functions in the cell nucleus and is widely involved in gene expression regulation, chromatin structure modification, and innate immune response. SP100 protein binds DNA and histones through its domains to regulate gene transcriptional activity, especially playing an important role in cell differentiation and development. It contains an HSR domain, which is essential for nucleosome targeting and dimerization, and a PxVxL motif, which is required for interaction with the chromoshadow domain.
