Background Information
PAF15, also known as PCLAF/KIAA0101, located at 15q14, is a small nuclear protein that binds to PCNA. Its expression fluctuated with the cell cycle, with high expression in S/G2 phase and degradation in G1 phase. Anchoring replication fork by PIP motif, specifically stabilizing the synthesis of lag chain, and regulating S-phase checkpoint; Participate in cross-injury repair when DNA is damaged. The gene is abnormally overexpressed in various solid tumors, which drives tumor progress by disturbing genome stability, promoting proliferation and antagonizing apoptosis. The overexpression indicates poor prognosis, and it is a tumor prognosis marker and potential therapeutic target. There are reports of 15kDa bands and 25 kDa bands with ubiquitination modification (PMID: 41606318).
